Extinction coefficients predict how much light a protein absorbs:

  • Units: M⁻¹cm⁻¹ (molar extinction coefficient)
  • 280 nm: Aromatic amino acids (Trp, Tyr) + disulfide bonds
  • Beer-Lambert law: A = ε × c × l
  • Applications: Protein concentration determination
